چکیده

An earlier communication (1) dealt with the application of the carbobencoxy method to the synthesis of peptides of L-serine. This work has now been extended, and an examination has been made of the response of several serine peptides and peptide derivatives to the action of peptidases. Synthesis of New Serine Peptides-In the course of the present study, the following new peptides were made: glycyl-L-serine, L-alanyl-L-serine, L-serylglycyl-L-glutamic acid, L-seryl-L-alanyl-L-glutamic acid, and L-serylglycyl-L-serine. In addition, several derivatives of these peptides are described in the experimental section of this communication. For the preparation of these compounds, the methods used previously (1) in the synthesis of L-serylglycine, L-seryl-L-alanine, and L-seryl-L-serine were modified only slightly. It was noted earlier (1) that the synthesis of seryl peptides by means of the reaction of carbobenzoxy-L-serinazide with the ester of an amino acid (or peptide) may be attended by the formation of 4-carbobenzoxyaminooxazolidone-2 as a by-product, thus reducing appreciably the yield of the desired peptide ester. In order to diminish the extent of this side reaction, the syntheses involving the use of the azide were conducted in a cold room at 4”. In the saponification of the carbobenzoxypeptide esters to the corresponding carbobenzoxypeptides, the yields were frequently unsatisfactory when the reaction was conducted in the usual manner; i.e., the treatment of the ester with 1.1 equivalents of alkali in methanol (or acetone) at room temperature for about 20 minutes, followed by acidification of the reaction mixture. With several of the serine-containing carbobenzoxypeptide esters, it was found by means of paper chromatography that the acidified filtrate, after removal of the expected carbobenzoxypeptide, contained appreciable quantities of ninhydrin-reactive substances. This suggests that

Synthesis of peptides of methionine and their cleavage by proteolytic enzymes.

In a previous communication from this laboratory (l), a method was described for the enzymatic resolution of Dr.,-methionine. The ready availability of the optically active enantiomorphs has made possible the application of the carbobenzoxy method to the synthesis of peptides of Land n-methionine. A search of the literature has indicated that the only methionine peptide described thus far is gl...

Antimicrobial Peptides Derived from Milk: A Review

Milk proteins provide a natural source of bioactive peptides with potential health benefits and applications in the food industry. The release of these peptides from milk proteins is achieved either by hydrolysis using digestive proteases or by lactic acid bacteria fermentation. Peptides, particularly those derived from milk proteins, can exert a wide range of nutritional, functional and biolog...

Applications of Zymography (Substrate-SDS-PAGE) for Peptidase Screening in a Post-Genomic Era

Peptidases are enzymes that catalyze the hydrolysis of peptide bonds in proteins or peptides. The hydrolysis can be specific or unspecific, leading to highly regulated cleavage of specific peptide bonds, or to complete degradation of proteins to oligopeptides and/or amino acids. Peptidases can be classified as endoor exopeptidases, the latter only act near the ends of the polypeptide chain. End...

Application of enzymes for the synthesis of the Cholecystokinin Pentapeptide (CCK-5) A contribution to Green chemistry

The reversal of the proteolytic cleavage of the peptide bond by proteases leads to the enzymatic peptide synthesis. In contrast to the chemical synthesis enzymes form the peptide bond without the danger of racemisation. The coupling can be conducted in aqueous buffer, no hazardous chemicals are necessary. Quite often side chain protection of the amino acids can be omitted, thus simplifying the ...

Protease- and acid-catalyzed labeling workflows employing (18)O-enriched water.

Stable isotopes are essential tools in biological mass spectrometry. Historically, (18)O-stable isotopes have been extensively used to study the catalytic mechanisms of proteolytic enzymes(1-3). With the advent of mass spectrometry-based proteomics, the enzymatically-catalyzed incorporation of (18)O-atoms from stable isotopically enriched water has become a popular method to quantitatively comp...
